I don't see a plotter selected in the top left. Is this a new setup or did this just happen out of the blue?

I have 3 orders of 170 full color 3½ circles. I set them up in illustrator 2 and save. When I place the registration marks and then attempt to save and it doesn't let me. I have both the labels & the paths on same page and when I attempt to save it, it wont save. What am I doing wrong?
