Don't confine your approach to any reference without a compelling reason. Just as sfr says, your 100% patch is your reference. If you really want to obsess, measure down the ramp from 100% until you find the highest C, then establish that as your reference.

That's a huge difference. I'm gambling that the sides somehow had different color management applied to them. Did anything differ in the process from creation/prep or the path through the RIP?

Looks like it may be a feed issue. Is the roll cycling slack then tension as it feeds? If it has a chance to feed one pass with the weight of the roll, then the next pass without that same back pressure, you can get this appearance.
